I will be in Venice in 1st week of April and then spending a day and a half in Florence before travelling to Le Sirenuse in Positano. What is the best way to travel from Florence to Positano? Which train and should we go thru Rome or Naples?

You could rent a car and drive down, maybe overnighting somewhere on the way down to break up the trip. A second option is to book a car service from Naples to Positano direct.
